Can I mix all the expansions or do I have to play one expansion at a time with the base game?

You can indeed mix all the expansions together if you like. They'll add extra strategy and difficulty to it. If you're playing for the first time however, I suggest playing the base game by itself first to get a handle on the basics.

Do you need the original game or is this a standalone game?

You will need a copy of the original game to be able to use this expansion. None of the other expansions are required, but it is compatible with any combination of them, too.

Does this support the six player expansion?

Yes, it does! With that expansion, you can play life up to 6 players, too. To be clear, the 5-6 Player expansion is not required to use the Life expansion.
